LMs w/ Activating KRAS Mutations Impair Lymphatic Valve Dev. Through Matrix Metalloproteinases

In this compelling research seminar, Diandra Mastrogiacomo, a graduate researcher at the University of South Florida, presents her work on the mechanisms underlying lymphatic valve loss in KRAS-driven lymphatic malformations. Lymphatic valves are critical for proper lymph flow, and their disruption contributes to severe complications such as chylothorax.

Diandra’s presentation explores how KRAS G12D mutations result in the regression and failed development of lymphatic valves by upregulating matrix metalloproteinases (MMPs) and components of the plasminogen activator pathway—culminating in extracellular matrix degradation. Using genetically engineered mouse models and 3D culture systems, she investigates potential therapeutic rescue strategies including MMP inhibition.

This talk offers key insights into the downstream consequences of hyperactive MAPK signaling in the lymphatic system and highlights promising directions for pharmacological intervention in life-threatening lymphatic anomalies.
